Scientific approach and technology
The Company has built a powerful technology platform to isolate antibody candidates to targets with challenging specificities. For example antibodies which distinguish between different receptor binding sites on the same ligand, recognise complex ligand and receptor or are capable of inhibiting ligand binding to complex multi spanning receptors on the surface of cells such as GPCRs (G-protein coupled receptors).
MBAS
Molecule Based Antibody Screening (MBASTM) – fast, effective and well proven for excellent functionality against a large number of different antibody targets, including small molecule drug, hormones, growth factors, simple receptors, carbohydrates and toxins.
CBAS
Cell Based Antibody Selection (CBASTM) – an innovative state-of-the-art technology platform that identifies antibody drug leads that bind to cell surface receptors in their natural environment, especially important for the discovery of antibodies to complex cell receptors such as G-protein coupled receptors (GPCRs) an important class of drug targets. Affitech has used the CBAS technology to generate antibodies to multiple different G-protein coupled receptors (GPCRs). GPCRs are implicated in the pathology of both inflammatory conditions and cancers.
Antibody libraries
Affitech A/S has a naive antibody-phagemid library with a diversity of about 10 billion different molecules. The library displays all V-families on the pIII subunit of the phagemid. It is made with an optimised packaging protocol, and the quality has been demonstrated e.g. by repeatedly isolating antibodies against a wide range of different target molecules (from haptens and lipids to GPCR membrane proteins).
In addition Affitech A/S has considerable experience in building and screening of more focused secondary libraries in a soluble scFv format for maturation of initial antibody candidates.
