Phage Display
Phage display mimics the fundamental steps of in vivo immunization offering the possibility of exploiting human antibody repertoires. Affitech’s library program generates human antibody repertoires of any desired isotype, e.g. naïve (IgM and IgD) and patient (IgG or IgE/IgA) repertoires.
The primary products of the antibody repertoire are the genes and the antibody fragments they encode. Such human antibodies are therefore directly accessible to methods of protein engineering, e.g. enhancing binding properties or altering any associated effector function.
Optimization of lead candidates
The lead candidates obtained from Phage Display are used as templates for a limited and directed randomization, generating secondary libraries. Robotic high throughput screening techniques are used to isolate antibodies superior to the original lead candidates from these libraries.
Depending on the specific demands, the optimization may e.g. increase overall affinity, stability or reduce unwanted cross-reactions.
